| - Food intake is motivated and initiated with the cooperation of internal informations regarding energetic storage of the organism and external informations allowing to give clues about the disponibility of food and its energetic composition. We chose to focus on the olfactory bulb (ob) which is the first center for food-related odors integration. It's sensitivity is modulated by the nutritionnel status of the organism, therefore acting as an important interface able to coordinate both internal and external informations, ultimately maintaining the energy homoestasis. During this thesis, we chose to target the olfactory-bulbar prokineticin 2 (pk2). This protein is highly involved in the transmission of the circadian rhythm and has recently been characterized as an anorectic hypothalamic neuropeptide. Moreover, we highlighted that the olfactory-bulbar pk2 expression was modulated by both the nutritionnal (fed or fasted) and the metabolic (lean or obese) status of animals. Using pharmacological and genetic tools, we have been able to emphasize that the acute injection of recombinant protein within the ob can significantly decrease food intake in mice, with or without leptin. Long-term approaches allowed us to demonstrate that the chronic down-regulation using a shrna can increase animals' food intake and that its over-expression inhibits it.
- La prise alimentaire est motivée et initiée grâce à la coopération d'informations internes renseignant sur l'état des réserves de l'organisme, et des informations externes permettant de donner des informations sur la disponibilité de la nourriture ainsi que sur sa composition énergétique. Nous avons choisi de nous concentrer sur le bulbe olfactif (BO) qui est le premier centre de traitement des odeurs associées à la nourriture, et dont la sensibilité est modulée par le statut nutritionnel de l'organisme, représentant ainsi une véritable interface coordonnant les informations internes et externes dans le but de maintenir l'homéostasie energetique. Au cours de cette thèse, nous avons choisi de cibler la prokinéticine-2 (pk2) olfacto-bulbaire. Cette protéine est fortement impliquée dans la transmission du rythme circadien et a récemment été caractérisée comme étant un neuropeptide anorexigène au sein de l'hypothalamus. De plus, nous avons mis en évidence que l'expression de la pk2 olfacto-bulbaire était modulée à la fois par le statut nutritionnel (nourri ou à jeun) et par le statut métabolique (mince ou obèse) des animaux. A l'aide d'outils pharmacologiques et génétiques, nous avons ainsi pu mettre en évidence que l'injection de protéine recombinante au sein du BO permet de diminuer significativement la prise alimentaire des souris, et ce, même en absence de leptine. Les approches à long-terme ont permis de mettre en évidence que sa sous-expression chronique à l'aide d'un shrna permet d'augmenter la prise alimentaire des animaux et qu'a contrario, sa sur-expression inhibe la prise alimentaire dans nos modèles murins.
