| - First, an innovating experimental methodology was applied for the optimization of the separation of nine psychotropic drugs by high pressure liquid chromatography. The modelling of the selectivity enabled us to obtain the best parting conditions for a standard analysis of drugs, with quantitative determination and satisfactory time of analysis. Then, the enantiomeric separation of a new synthesized aryl propionic acid of pharmaceutical interest (anti-inflammatory) was optimized by capillary electrophoresis. The desirability function approach has been used in order to optimize simultaneously the resolution and the migration times of both enantiomers. Finally, the validation of a rapid and sensitive HPLC method for routine control of oxaliplatin in a formulation (Eloxatin®) has been reported. An experimental design was used to assess the robustness and the precision of the method.
- Une méthodologie expérimentale innovante a d'abord été appliquée à l'optimisation de la séparation de 9 psychotropes par chromatographie liquide haute pression. La modélisation de la sélectivité a permis de retenir des conditions de séparation optimales pour une analyse de routine, avec une détermination quantitative des solutés et un temps d'analyse satisfaisant. Ensuite, la séparation énantiomérique de l'acide aryl propionique présentant un intérêt pharmaceutique (anti-inflammatoire) a été optimisée par électrophorèse capillaire. L'approche de la fonction de désirabilité a été utilisée pour optimiser simultanément la résolution et le temps de migration des deux énantiomères. Finalement, la validation d'une méthode HPLC rapide et sensible pour le contrôle standard de l'oxaliplatine dans une formulation Eloxatin® a été décrite pour évaluer la robustesse et la précision de la méthode.
