| - Depuis de nombreuses années, les réactions de couplage pallado-catalysées ont profondément révolutionné le domaine de la synthèse organique. Plus récemment, de nouvelles méthodes reposant sur la fonctionnalisation directe de liaisons C-H de noyaux aromatiques ont suscité un intérêt tout particulier. Cette méthodologie permet désormais la formation régiosélective de liaisons C-C, C-N, C-O et C-X (X : halogène) à partir de matières premières faciles d'accès ne nécessitant pas de fonctionnalisation préalable. Dans le cadre de ces travaux de thèse, nous avons développé des méthodes de fonctionnalisation C-H pallado-catalysées de benzamides en mettant en jeu un nouveau groupement ortho-directeur : le N-acylsulfonamide. Ce dernier s'est révélé très efficace et peut également subir diverses transformations chimiques à l'issue de la réaction de fonctionnalisation C-H. Dans un premier temps, une méthode d'arylation de N-tosylbenzamides a été mise au point. La transformation fonctionnelle du groupement directeur a ensuite été étudiée et a notamment permis d'accéder, par amination C-H, à la structure phénanthridinone précurseur de ligands des récepteurs sérotoninergiques 5-HT4 synthétisés au laboratoire. Dans un second temps, des méthodes d'alkoxylation et d'halogénation C-H à température ambiante ont été développées, avec ce même groupement directeur, et ont permis la synthèse de N-tosylbenzamides diversement ortho-fonctionnalisés. La dernière partie de ces travaux a été consacrée à la conception et à l'évaluation de nouveaux ligands fluorés des récepteurs sérotoninergiques 5-HT4. Cette étude a permis de mettre en évidence l'influence de l'introduction d'un atome de fluor sur l'affinité de ces ligands.
- Over the past four decades, palladium-catalyzed cross-coupling reactions have profoundly revolutionized organic chemistry. More recently, transition-metal-catalyzed C-H bond functionalization of arenes has emerged as a new and promising methodology. This process allows the regioselective formation of C-C, C-N, C-O and C-X (X : halogen) bonds from more easily accessible starting materials precluding prior activation. As part of this PhD project, we used the N-tosylcarboxamide group as a versatile and transformable ortho-directing group to achieve palladium-catalyzed C-H bond functionalizations of arenes. In the first part, we developed C(sp2)-H arylation method of benzamides. The functional transformations of the N-tosylcarboxamide group were then accomplished including the synthesis of phenanthridinone structure, by C-H amination, which is a precursor of serotoninergic 5-HT4 receptors ligands synthesized in the laboratory. In the second part, the room temperature ortho-alkoxylation and -halogenation were performed using this directing group leading to the preparation of a wide variety of diversely ortho-functionalized N-tosylbenzamides. The last part of this project focused on the design and biological evaluation of new fluorinated 5-HT4 receptors ligands. This study highlighted the influence of the introduction of fluorine atom on the affinity of these ligands towards 5-HT4 receptors.
